ALTHEA Gold™ Antibody Library Kit

Technical Description

The ALTHEA Gold™ Antibody Libraries are advanced semi-synthetic human antibody phage display libraries for therapeutic antibody discovery (1). The libraries are built on two framework combinations, VK3-20/VH3-23 and VK-4-1/VH3-23, to modulate the shape of the paratope and enable the recognition of a wide range of potential protein & peptide targets. Sequence diversity is concentrated specifically in the CDRs, at key antibody-antigen interaction sites that were identified through comprehensive analysis of known antibody-antigen structures, except for the CDR-H3, which are of human origin and derived from a large pool of donors. The synthetic primary libraries in scFv format were exposed to a heat-shock and rescued by protien A to eliminate unfit cones and select for thermostable, developable antibodies (Fig. 1). A subsequent shuffling using H3J fragments derived from human donors resulted in a 100-fold increase in diversity and a 50% increase in thermostability. Each library contains more than 10 billions unique clones..
