The ALTHEA Gold Libraries™ were developed by Antibody Design Labs in the US in 2018.
A Unique Construction Process
An innovative 3-step thermo-filtration strategy eliminates unfit clones and selects for thermostable, developable antibodies.
A Carefully Designed Diversity
Sequence diversity is concentrated specifically in the CDRs, at key antibody-antigen interaction sites that were identified through comprehensive analysis of known antibody-antigen structures. A subsequent shuffling using H3J fragments derived from human donors resulted in a 100-fold increase in diversity and a 50% increase in thermostability.
A Dual Frameworks For Antigen Recognition
The antibody’s ability to recognize antigens relies on its binding site, called the paratope. The ALTHEA Gold libraries™ utilize two different framework combinations, VH3-23/VL3-20 and VH3-23/VL34-01, to modulate the shape of the paratope and enable the recognition of a wide range of potential protein & peptide targets.
Antibodies to Any Targets!
A typical screening of the ALTHEA Gold libraries™ leads to multiple binders in only 3 rounds of panning. Additiontal rounds of solution capture identifies the best binders. Cross-selection identify reactive clones to the cynomolgus and murine homologs.
